Clinical Pharmacology
Aciclovir is a synthetic purine neucleotide analogue, which acts against human herpes viruses. The enzyme thymidine kinase (TK)encoded by HSV, VZV, and EBV converts Aciclovir into Aciclovir monophosphate, a nucleotide analog, which is further converted to diphosphate and triphosphate by cellular enzymes. Aciclovir triphosphate interferes with DNA polymerase and inhibits viral DNA replication with resultant chain termination following its incorporation into the growing viral DNA chain.
